I'm still having this issue. I Adopt a boy and girl animal, put them into an exhibit and they say they can't mate because there betas. In the past someone said it's because in sandbox if you have animal fighting turned off it will cause this to happen, i have fighting on. I did make a post on the issue tracker a long time ago. I try sending them to the trade center and back, i also tried getting another animal to see if that'd help.

Do you have any other wolves in the habitat or in your Zoo?
In wolf packs only the Alpha pairs mate. So, in game this would be the same. Only your Alpha pair of wolves will produce any offspring. If you want to have a new alpha pair you will need to remove your current pair from the Zoo. For example, in my Zoo my alpha female passed away due to old age and her daughter took her place. But that left her dad still as the Alpha male. So, I had to release him to the wild in order to get a new alpha male for her to mate with.
