Maybe I just haven't come across this yet, but for passenger missions, what I see available are tourist trips. The passenger always wants to be returned to the same port. But one thing I haven't seen is passengers wanting to be dropped off at another location, and not wanting to see sites. I think having a mission where you deliver someone somewhere else would be fun. I've also recently been doing chained missions which have a lot of potential, I think.
The way this could work is you accept passenger mission to destination A...once you drop them off at A, a chained passenger mission would be to the next stop. If enough stops close a loop, back to the starting station, then that's a circular bus route. This would make sense if trade routes that may exist, etc.
