I'm thinking about getting a Vkb gladiator und using it alongside my TWCS throttle. Is the best way to do this is to use Vjoy to combine the axis+

Not sure what you're asking. I used a VKB gladiator MkII and a TWCS for a while before I upgraded the throttle to a Virpil one and it was just like setting up any HOTAS. Currently, I use a Contellation Alpha, VKB Gladiator-K, and Virpil throttle and set up was pretty straighforward. I don't use Vjoy though, I always used JoyToKey. Elite recognizes all the analog axes natively with no problem and the software lets me remap the buttons to keypresses.
Yep, I use a VKB Gunfighter and the TWCS throttle. Just assign the axis that I want in controls, no need to use any intermediate software.
