What's the best way to plot a route to Tir or even Colonia.

Have a google search for the Colonia highway. Breaks it down into nice 5 kly sections.

If you wish to move a bit away from the overly explored Colonia Transit Corridor (i.e. to make 1st Discoveries), mark yourself some Waypoints anywhere in the Galaxy Map and feed those (or known Systems in their proximity if the individual System isn't in EDDN) after another into the Spansh Route Plotter.
