I've always loved the Evil Dead series (the originals)

Great blend of humour and Bruce Campbell. :D

Indeed - LoB is a tough one to beat.

Another classic would be Airplane!

So many quotable lines and the late great Leslie Nielsen.

South Park Bigger longer and uncut. the funniest musical I've ever seen.

South Park. Good call. But where there's a South Park there's a Team America. There are so many wonderful lines I can't quote here but I think this one will make it through:-

I just looked through my collection and there is no way I can choose the best, there are just so many genius comedies.
I will mention Burn After Reading though, just because it did have many laugh out loud moments and it is probably overlooked.

Also I will unsuccessfully restrain myself from mentioning Dr Strangelove...
